Notch信號靶標PCR芯片可用于研究Notch信號轉(zhuǎn)導的84個關(guān)鍵應(yīng)答基因的表達。Notch信號通路是一個傳統(tǒng)的發(fā)育途徑,參與細胞間通訊,細胞命運,細胞凋亡和發(fā)育。Notch信號通路首先在果蠅中發(fā)現(xiàn),哺乳動物同源的Notch信號通路稍晚些被確定。來自Delta及Jagged/Serrate的配體可以激活與膜結(jié)合的Notch受體,誘導Notch胞內(nèi)結(jié)構(gòu)域斷裂。這種胞內(nèi)結(jié)構(gòu)域與RBPJ家族的轉(zhuǎn)錄因子以及共激活因子和輔阻遏物的相互作用來啟動靶基因的轉(zhuǎn)錄。Notch信號通路激活的輸出依賴于細胞內(nèi)容物,在適當?shù)臅r空調(diào)節(jié)激活是正常的胚胎發(fā)育過程所必須的。Notch信號通路失調(diào)可能會導致癌癥以及遺傳發(fā)育障礙。通過如ChIP和基因表達研究等實驗技術(shù),很多經(jīng)典的Notch通路靶標基因都已經(jīng)被鑒定出來了。該芯片包含了Notch經(jīng)典信號通路的轉(zhuǎn)錄因子和研究驗證得到的高關(guān)聯(lián)性靶標基因,通過該芯片得到的結(jié)果可用于Notch信號傳導激活和抑制研究。通過實時定量PCR的方法,研究者即能夠利用該芯片簡單可靠地同時檢測Notch調(diào)節(jié)的細胞活動相關(guān)基因的表達。
