凋亡PCR芯片可用于研究參與細(xì)胞程序性死亡的84個關(guān)鍵基因的表達(dá)。細(xì)胞凋亡在正常生物過程中發(fā)揮重要作用,如分化、發(fā)育和體內(nèi)平衡。應(yīng)激反應(yīng),如熱休克、缺血、非折疊蛋白和病毒感染,會引起細(xì)胞嚴(yán)重受損甚至細(xì)胞凋亡。細(xì)胞凋亡經(jīng)常導(dǎo)致不可控的細(xì)胞生長,如腫瘤形成和癌癥;同時腫瘤壞死因子(TNF)受體也會通過激活FADD和其他死亡受體蛋白引起凋亡。這一芯片包含了TNF配體及受體、bcl-2、半胱天冬酶、IAP、TRAF、CARD、死亡域、死亡效應(yīng)域、CIDE家族,以及參與p53和DNA損傷通路的基因。研究這些基因的表達(dá)有助于確定在生物模型和細(xì)胞模型中的細(xì)胞機制。通過實時定量PCR的方法,研究者即能夠利用該芯片簡單可靠地同時檢測細(xì)胞凋亡相關(guān)基因的表達(dá)。
